Self-diffusion of reversibly aggregating spheres.

Reversible diffusion limited cluster aggregation of hard spheres with rigid bonds was simulated and the self-diffusion coefficient was determined for equilibrated systems. The effect of increasing attraction strength was determined for systems at different volume fractions and different interaction ranges. It was found that the slowing down of the diffusion coefficient due to crowding is decoup...

The behavior of horse liver alcohol dehydrogenase in guanidine hydrochloride solutions.

The z-average molecular weight of horse liver alcohol dehydrogenase in dilute neutral buffer is 78,200 by sedimentation equilibrium. Guanidine hydrochloride, 1 M, reversibly inhibits enzymatic activity. The inhibition with nicotinamide adenine dinucleotide is competitive and with ethanol is mixed. When ethylenediaminetetraacetate is added to the enzyme in 1 M guanidine hydrochloride, zinc is re...

A comparison of weight average and direct boundary fitting of sedimentation velocity data for indefinite polymerizing systems.

Analysis of sedimentation velocity data for indefinite self-associating systems is often achieved by fitting of weight average sedimentation coefficients (s(20,w)) However, this method discriminates poorly between alternative models of association and is biased by the presence of inactive monomers and irreversible aggregates. Therefore, a more robust method for extracting the binding constants ...

Sedimentation of Reversibly Interacting Macromolecules with Changes in Fluorescence Quantum Yield.

Sedimentation velocity analytical ultracentrifugation with fluorescence detection has emerged as a powerful method for the study of interacting systems of macromolecules. It combines picomolar sensitivity with high hydrodynamic resolution, and can be carried out with photoswitchable fluorophores for multicomponent discrimination, to determine the stoichiometry, affinity, and shape of macromolec...

Streptolysin O: sedimentation coefficient and molecular weight determinations.

The sedimentation coefficient of streptolysin O as determined by sucrose density gradient ultracentrifugation is 3.7S. An approximate molecular weight of 60,500 was calculated from the sedimentation velocity, and a similar value was obtained by Sephadex gel filtration. There was no measurable difference in the sedimentation coefficient of streptolysin O in either the active or reversibly inacti...
